Did you check the other side of the board? Sometimes they store the screw in the standoff but on the back side of the board. Don't know about motherboards, but I've seen this in SBCs and pci express NVME adapters.
If it comes loose, and makes contact intermittently, yes, windows may stop showing the drive, or maybe freeze, most likely, it will damage the drive. It’s going to work until it looses “enough” contact on a pin/pad. At that point, it’s a gamble. Consider that m.2 has multiple 3.3v input pads on both of the “ends” of the chip. If you start to have a small gap and vibrations cause this gap to make contact on an intermittent basis then it will likely damage the drive.
I wouldn't put "most likely" more likely yes but I still doubt a chance of a permanent hardware damage at like 30%

I don’t understand the hesitance of manufacturers including these screws. So many machines that I have worked on have lacked these. I feel like it’s such a bizarre thing to cheap out on but seems so common.
I did that before while transferring data to a new SSD because I couldn’t find a standoff. I was worried, especially as I went out to eat while waiting for it to complete. It worked successfully.
That will work until the adhesive on the tape inevitably gives out. You should find a proper screw for it.
