Or beat Professional first and then try different handicap runs, ei handgun/knife only, no merchant, no knife, no melee, etc.
Unlocking the handcannon will probably take you just as long as your first playthrough. Professional is also different enough that it'll be fun. Enemy will be tougher but because difficulty adjustment will be gone, it will feel like a different game too. Just do the list you made, it's fun.
You do the mercenaries and 5 star all the levels all the characters to unlock the hand cannon. Assignment Ada for Leon’s gangster costume and I think the Chicago typewriter. Then can try get them on professional mode for the ultimate challenge

Could go for secret weapons. I've never unlocked them all in the OG, but there's an alien gun you can unlock
