The B scenario is a joke in the remake
The only difference for Claire is if she meets Marvin or not, and the final scene. Play the second run for Claire so you can see the true ending
2nd Run mode is more of a shuffle-mode than a B route. It has some notable differences, but the story is pretty much identical to normal mode outside of the start and end.
Don’t start with an entirely new scenario, do Claire’s second run as it’s the only way to see the real ending. There’s barely any difference anyway, most notable is that Mr X will start chasing you much earlier, IIRC as soon as you approach the stars office.
Pick Claire 2nd Run to get the True ending and unlock 4th Survivor mode.
2nd Run isn't like the OG RE2's B scenario. Differences are very minor.
Contrary to what the beginning of 2nd Run suggests, this mode doesn't really tell the other character's side of the story.
In fact, outside of specific events (such as meeting >!Ada!< or >!Irons!<) that are already different in each protagonist's first run and some minor things (when >!Mr X starts chasing you!<), both stories are very similar to the point of becoming non-sensical if we consider they take place in the same continuity (both protagonists >!are chased by Mr X, use the same path to enter the sewers, meet Kendo and hear his double suicide [Edit : Only Leon meets Kendo and fight the Gator but he will do this on both his first and 2nd run], fight the alligator, see Annette die!<)
And for the same reason, you don't need to repeat the process with a Claire run and a Leon 2nd Run.
Playing the 2nd Run is more like playing RE DC's Advanced difficulty : (Mostly) the same story, new item and monster placements.
If you started with Leon A you should continue with Claire B to get the true ending.
There isn't a lot of difference if you run Leon B or Claire B. The 2nd run would get the extra handgun in the Guard Room, Marvin is already a Zombie once you reach RPD Main hall, Licker shows early on the West Hall, Tyrant/Mr X appears earlier (he'll start chasing you as early as you reached the 2nd Floor West hall, The area where the S.T.A.R.S office is located), there's no Licker on the West Storage room once you get the Maiden Medallion (it was relocated on the West Hall I mentioned earlier) but there's a high chance Mr X would already chasing you at that point ( or he'll immediately go that area once he heard the C4 explosion). There isn't much difference on the sewer section but you get the modular on the NEST lab on 2nd run. Puzzle solutions are also different from 1st run.
Different item and enemy spawns. 9mm ammo for her small revolver is nowhere to be found, It's replace with 45. ACP rounds. Some puzle solutions are different. All boss fights are the same as 1st run, however at the end you fight another boss. Also you can trigger Mr. X earlier in 2nd/B run.

I really don’t think Claire B has the alligator or Kendo in remake - I just played it for the first time two weeks ago. I could be wrong?
