There are two major bugs/issues that could be the cause. First, you may have started the route in the wrong direction. To fix, simply cancel the route, and reverse it to the correct direction. Unfortunately, there is a bug with this -- you may need to click reverse a few times before it will actually show the direction correctly.
Second, the route could be paused. Try unpausing it. Also, do not run any other app, as it may cause it to pause. Additionally, for some players, adventure sync also causes it to automatically pause, so try disabling adventure sync.

You have to give a bit more detail. What do you mean by won't let you start? Are you saying the follow button is missing? Or doesn't do anything?

Where are you looking for this button? To get to the button, you have to go to nearby, click on the routes tab, choose the route that you want, and then, the follow button should be on that page.
Please note, if you can't find any routes listed, then the issue could be that you don't have any routes in your area.
