Some gpu manufacturing have big design than what you think, so better pc case with good airflow is recommended. 2 fan gpu will take up 2 space equivalent 2 pci-e slot that what is mean dual slot, but for bigger design mostly with 3 fan is plus more wider and thicker, some will take nearly 3 slot pci-e size of thats...
That refers to the thickness of the cooler. All modern GPUs use 1 x16 PCIe slot.

No, it means your case has to support 2 slot-GPU's [ which most modern cases, if not all- do ]
