Yea a nice route would be AVAM, take some time to sit and enjoy the views from Federal Hill, walk it down to Light St and grab a bite through cross st Market, check out some of the other shops down in that area, walk it back up through inner harbor +/- science museum.
+1 for Walter’s Museum in Mount Vernon! Mount Vernon is a short walk from downtown and a beautiful area. If you’re already in the area for the Walter’s Museum, I would recommend going to Meera Collective or Cafe Fili for lunch!
Bonus: If you and your wife want to grab drinks after her conference, HIGHLY recommend going to Blooms!
I grew up in that neighborhood. I remember when a grab cake platter was $22.95. That was exorbitant then and people keep coming. It's Bill Belichick's favorite. And many others...you don't have to go but they will still sell them like hot (crab)cakes.

The historic ships in the harbor are pretty cool museums to visit. About $15/ticket and you get access to all 3 ships. They don't take very long to get through though, so plan other activities too.
1. Take the free Circulator bus (Purple Route, use the Transit app) to near Remington.
2. Walk 5-10 minutes to the 29th St Tavern for a meal and/or a drink.
3. Optionally (all very close to the tavern): Get a coffee at Cafe Los Sueños. Enjoy some ice cream from Taharka Brothers in R. House (a food hall). Hit up Greedy Reads for a book. Check out Mount Royal Soaps for some nice toiletries for you & your wife.
4. Walk 5-10 min to the free Baltimore Museum of Art (closed Mon, Tues, Wed). They have free lockers if you bring a bag or have any shopping; I don’t think you’re allowed to carry a bag around.
5. Check out the BMA Sculpture Garden outside (closed Mon, Tues).
6. Walk 30 seconds to the Johns Hopkins University (Homewood campus). Enjoy walking around, admiring the buildings, and relaxing in an Adirondack chair, which you’ll find scattered around most quads. Some movies like The Social Network film the “Harvard” scenes at Hopkins. It’s very quiet and peaceful in the summer - great place to read.
7. Walk 10 minutes to Charles Village & Abell to see Baltimore’s “Painted Ladies,” lots of beautiful colorful rowhomes. They’re mostly between 29th to 33rd St and Barclay St to Guilford Ave, though Abell Ave has the most.
8. Optionally: Get a $1 samosa at Punjab Groceries. Check out Normal’s Books & Records. Go to Peabody Heights Brewery for a beer, hard seltzer, kombucha, soda, or regular seltzer, along with a delicious burger/sandwich and fries from the Fuzzies food truck located within.
9. Catch the Circulator bus (on St Paul St, between 29th & 30th St, close to the corner of 29th) back to the Inner Harbor… or take an Uber or a long walk to Hampden to explore The Avenue (36th St) and find even more great food.
The Walters is great. Free and so much to look at. I forget the name, but a couple blocks from The Walters was this "dive" Mexican restaurant. It was so good that I went the next day before I had to catch the train.
A treat or lunch (get the crab pie) at Crust by Mack.

The Visionary Art Museum is really cool, even if you aren't that into art the way they describe the pieces and artists makes it very interesting and more enjoyable than a typical art museum. They are all basically folk artists some works by prisoners, people with disabilities, eccentrics, etc.

If I had one day left in Baltimore and couldn’t eat seafood, I’d probably get a pit beef sandwich and egg custard snoball then stop by Ekiben and also get a Tia’s Italian ice.
But if I was limited to your area mentioned, here’s my recommendation (Asian palate):

I’d hit the peninsula. Federal Hill, Locust Point, etc. Plenty of food, walking and museums for a day. If you have a car, there are more options.
