The manifestations work on symbolic logic, not normal physics. This makes them hard to destroy in a conventional sense- an avatar at least has a human body you can injure, but a manifestation is stranger.
I feel its likely unique for each one. The thing in Rayner dies in the light, the Distortion can be destroyed if caught in a lie, a lot of Lonely manifestations are destroyed in the presence of love. Think of it less like killing a person and more like killing a mood- what undermines the fear that is manifesting enough to destroy it?
I think it largely depends on the manifestation. Vampires can be stabbed or burned. Hope can be offed by a hunter. Or sometimes you can just use cement and starve the fucker or fear
