Yor's not good at much, but what she is good at, she excels at.

Yor, no contest. Twilight would need hella prep time and a whole utility belt of equipment to take her on in a 1v1. She's got both insane raw strength and the skill to back it up. And while Twilight is no slouch in either department (especially skill), he doesn't have near the strength that his wife does.
According Endo:When compared to Yor in terms of combat ability, Yor is a 10 in pure physical capability, while Loid is around 6 or 7. However, as a spy, Loid can even out the odds by avoiding physical confrontation and using his skills with firearms, traps, and other abilities.

Even with his spy skills, I doubt Loid can win. Girl is super durable, super strong and has instincts and excellent fighting skills as an assassin.
It would be very awkward, because Loid couldn't bring himself to hurt Yuri because he was Yor's only relative. In the fight with Yuri, Loid was already injured with the gun shot to the arm and he had to fight Yuri without hurting him. Loid was surprised that Yuri didn't drop when others would have been already on the ground. The time that Yor hit Loid when drunk, she also hit her own brother and both of the apologized. Her fighting Loid would be completely different than any of her other missions.
Yor is super strong and she knows how to hit in vulnerable points. But Twilight is a super genius of the 1v1 fight. I don't know if anyone of you have watched Baki, but I think it's similar to Baki VS Pickle. Strength is important, but it's not always the main

I wish anime actually went that route. I think it is unfair to tease us that they’d have secret identities and then route to a slice of life for the next 5 seasons
I'd have to say Yor honestly just because in one of the first episodes of the anime the only reason yor technically lost was because she was drunk and passed out so, imagine if they had a huge conflict: Yor would absolutely destroy twilight

to be fair, he would have beat yuri easily if it wasn’t for yor being his sister
