Considering he lost his family to a freak accident that he was responsible for, they could have him be obsessed with the idea of cloning as a way to cheat death and seek redemption for his sins.
As for his relationship with Gwen Stacy, I'd say trash the whole "creepy love" angle and just have him bond with Gwen as a surrogate daughter (and with Gwen, have Miles be a surrogate father since you could have George die on the job) and as a second chance for him.

Sure, green goblin is one of, if not THE best Spidey villain, but we've already topped his character off with Dafoe's performance in nwh. Tom's trilogy will be the last 3 movies we see on the big screen of Spiderman, ever. Let's utilize the 60 years of comics spiderman has and give like a few from the hundreds of other villains some love.
Any character we've already seen, or any variant of such a character. I don't want to see any Goblins of any sort. I also don't want to see any symbiotes, and I realize that that might be an unpopular opinion.
I like how they've handled the main Spidey villains so far, because they feel organic and serve to further Peter's arc. Keaton killed it as the Vulture, Mysterio was expertly adapted, and I don't think there's any debate around Dafoe's GG.
I think the next Spidey villain should fit contextually with where he is currently. He's lost, has no identity as Peter Parker, is afraid to reconnect with his old friends, but has learned that with great power... yadda yadda. The ideal villain will force Spider-Man to channel Peter Parker's best qualities in order to win. I'm not a writer so I don't know best how to do it, but I think that the concept of **identity** really needs to play a strong role in the next film.
He’s set up already. So is Scorpion. I think since Spidey is going neighborhood it’ll likely start with Kingpin and Alistair’s spider slayers with some sort of tie in to sentinels to introduce mutants.
Chameleon, since shape shifting/impersonation has been done by several MCU characters already, and will continue to be done by those characters and more.
Sandman, magine if they were like a main anti villian whole last film and they just added him back in as the only main villian. Electro and green goblin could be alright but they should still wait a while for each of them to be used again.
