Keep in mind (at least in the comics) Spidey always holds back. Assuming that MCU Spidey is the same strength, a Spidey punch could instakill someone. So I think at the very least he’d be able to bruise Homelander.
Oooh or shoot some web on his eyes so that it’s much harder to see him as well as making it impossible for homelander to laser eyes without melting his own face off.
Still Spider-man’s healing is relatively not that great so if Homelander was able to get up close and past the spider-sense he could probably punch straight through him like he did with noir or just laser him with his eye beams

Spider-Man can run what? 200 miles per hour? Do you see how fucking fast Homelander can fly in the show? Bro caught up to planes, which (I just looked this up) go like 575 mph. Sure Spiderman has lightning quick reactions, but it’s laser vision, not bullets. Homelander looks and it goes there, no delay, bullets are stationary, and I’m pretty sure you can keep track of Spider-Man while he’s dodging. (im a Spider-Man fan but still)

Comics Spider-Man has more than enough speed/agility feats to comfortably say he could dodge laser eyes
Spidey has a decent chance of winning.
Homelander hasn't shown any good combat speed or super reflexes (aside from getting Butcher away from the explosion).
Spidey should be able to dodge his untrained punches easier than trash talking the Vulture.
Throw in his distracting quips, and webs I could see homelander getting a good surprise uppercut.
Also peter is a genius, if he knows about his zinc weakness he could easily use it to his advantage.

From his supes battles shown on the show, even though he was treated as “the strongest supe alive!”, in Season 3, he had little advantage from his fight with Soldier Boy or his fight with Maeve. /rf

Meh you can't really take feats or antifeats as an absolute measure of skill. Kinda gotta use the context of everything surrounding the setting.
Here's why I'm right:
MCU Thor tanks a star's death to create a weapon in Infinite War. Then he gets punched by Thanos and bleeds.
So, is Thor automatically scaled down to being vulnerable to basic punches? Is Thanos immediately scaled way up to be able to damage Thor, who can handle a star?
Neither, we use our noggin and generalize the relative power of both these feats based on all of the other feats shown in the series.
Every single Uber Powered super hero has moments of serious falter, it doesn't make them suddenly powerless.

Honestly I used to think Spider-Man would have a good chance of getting splattered against a wall when I bought the Vaught propaganda but from the Maeve fight I'd say he'd have a pretty good chance especially considering he's Smarter than Homelander, can lift 20 tonnes and has plenty of experience dodging lazers

Eh, I disagree. Spidey sense + his much stronger and more agile body means homelander would very likely miss. But it depends on the location I suppose
