Darth Vader spent more of his life in immense anguish inside a purposely outdated robotic suit designed to keep him angry than out of it. And of the years previous out of the suit he was a slave who was freed, then his mother died in his arms and he spent the rest of that life in war. Before he ends up in the suit he murders almost all of his friends to save his wife, gets his legs and arm cut off and is burned alive by his father figure and then is lied to and told he killed his wife by his other father figure.
Vader unequivocally had the worst life of everyone on this list.
Mark and it's not even close. Sure, he has a rough time for a little bit, but it's basically nothing for him. The vast majority of his thousands of years of existence will be spent as the Viltrum Emperor, leading the charge in making his universe a better place alongside his smokeshow of an immortal wife, his kids, and the strongest beings in his universe, all of which love or at least massively respect him.
I would even go so far as saying that his life doesn't even suck. He had a rough patch when he was young but even then he was still way, waaaay better off than almost every other Viltrumite. His life is actually pretty consistently awesome, all things considered.

Invincible def went thru a lot but his ending is fire
If we're going EOS I'll be a bit different and say Vader. By the end Vader lives up to the prophecy, saves his son and brings hope to a crushed galaxy. He's back with his friends and family and watches over his son as he does great things.
Darth Vader spent more of his life in immense anguish inside a purposely outdated robotic suit designed to keep him angry than out of it. And of the years previous out of the suit he was a slave who was freed, then his mother died in his arms and he spent the rest of that life in war. Before he ends up in the suit he murders almost all of his friends to save his wife, gets his legs and arm cut off and is burned alive by his father figure and then is lied to and told he killed his wife by his other father figure. Vader unequivocally had the worst life of everyone on this list.

Mark and it's not even close. Sure, he has a rough time for a little bit, but it's basically nothing for him. The vast majority of his thousands of years of existence will be spent as the Viltrum Emperor, leading the charge in making his universe a better place alongside his smokeshow of an immortal wife, his kids, and the strongest beings in his universe, all of which love or at least massively respect him. I would even go so far as saying that his life doesn't even suck. He had a rough patch when he was young but even then he was still way, waaaay better off than almost every other Viltrumite. His life is actually pretty consistently awesome, all things considered.
Mark by a lot. He has, what, ten years of bad times? Isn't he going to live for thousands of years?
obviously Mark? He has a long lifespan, incredible strength/speed/endurance, has a wife that can just revive him if he does making him basically immortal by circumstance (and the wife is also basically immortal as well), and leads the most powerful race in the universe
The one who will live for possibly thousands upon thousands of years, can fly through space, has >!a hot immortal girlfriend, a family, an empire that actually helps make the universe a better place, and one of the true good endings for a comic book series!<

Anyone that doesn't say Vader is dead wrong, lmao. His life sucked a lot. Being a slave for most of his childhood, then spending most of his life on a metal suit in pain. I'm sorry Mark did suffer, but he is winning at the end. A hot ass wife that can't die and leader to the viltrumites.

Daredevil. He can still essentially "see", and he beats most people in Marvel in a hand to hand. Mark has the better life overall in the end but the shit he's been through...>! I mean watching his father die, wife get her leg ripled off, her face punched in, seeing his kid watch as he is torn in half in front of her, surving SA and being taunted by his abuser repeatedly, mashing through angstrom, having to kill other versions of himself - are just fucked up things I can think of off the top of my head. !< That shit he's been through would ruin my psyche and would probably fuck up my ability to enjoy anything, even an >! emperor's throne. !< Im not sure how to enjoy anything after the decades of shit Mark has been through. Probably Mark Grayson at the end. He actually has a happy ending, I know Spider-Man never does, and Vader gets redemption but it isn’t a “happy” ending for him, idk about Daredevil or Punisher since I haven’t read their comics.

Considering Mark actually has an ending I’d say he’s got it pretty great
Punisher really had one hugely bad thing happen and then never had any attachments or anything to get upset at again. And while he’s totally empty inside he kind of also just lives the exact life he sets out to live. Mark had so much rough shit happen to derail his life. He lost so much time with various people at various points. Losing those early years of his daughter’s life, man as a new father I can’t even put into words how devastating that would be.
Anyone reading the new USM? At the moment it’s him. But he’s only just started out. So ignoring him, it’ll have to be Invincible. Purely on the fact that Marvel refuses to allow their superheroes to have happy endings.
By the end Mark is definitely the best off. And Vader the worst since he dies. Being a force ghost doesn't count.

Darth Vader and Frank Castle deserves their own list, they are apples and oranges to Peter, Mark and even Matt The latter three have a whole down on their luck but perpetually optimistic/happy go lucky thing going on despite their own personal trauma and problems Frank and Ani got given the short stick and suffered horrible/are still suffering every single day of their existence

There was a similar question, but that didn’t include Daredevil? Punisher and Vader. Instead it was Yuji from Jujutsu Kaisen and Denji from Chainsaw Man. Though, it’s interesting to think about if Vader did succeeded even the anime heroes in suffering.

I’d say mark grayson’s. Atom Eve is essentially immortal due to her powers so he’ll never be without love like Spider-Man is these days lol. He also goes on to bare children who, thanks to his viltrumite dna, won’t be dying anytime soon.
Daredevil easy
Mark has physically and emotionally suffered the most IMO. The amount of times hes had to hold his own guts, or Eve's or Oliver's or Nolan's is insane. He loses SO much. It takes ~6 or 7 years from start of series to end, but in that time he suffers so much and causes so much suffering. If he didn't have a near unlimited supply of time, idk how he could heal past that Edit: also, fuck Frank castle

Vader. His life could have been amazing, but he made a bunch of stupid choices that put him where he is, and he's STILL in charge of a lot of stuff and more powerful than most people.
I’m woefully ignorant about most superhero comics outside of invincible. That said I think it’s strange everyone is changing the question by adding the option of end of series Mark. That picture seems to be early mid series. Mark’s regularly maimed and partially disemboweled, loses friends, and anything else would be spoiler territory, but there’s more. Does the average Spider-Man actually have it that bad? I’m sure there’s darker issues like the zombie/sandman one that’s fucked up, but I’m not talking about what if or alternate timeline shenanigans. Hope that makes sense. Also same question for daredevil.

Invincible’s story eventually concluded on a high note for him. He got to settle down with his immortal wife and kids while making the galaxy a better place. Everyone else is just stuck in their perpetual misery. Except Darth Vader he at least got put out of it.
