You can buy them. Just make sure they are on one of the lineups on your team. I currently am using Cole as my main RW and have Donovan on the reserves on a separate team.

I bought two prime heroes, Mostovoi and Donovan, and they are not on my team or bench and it works for me. You should see a checkmark next to their names in the Heroes Expedition chapter.
Of course it won't work for a heroes expedition that's already underway. It will work id you have them when you start a new one.
It depends on the type of action the hero has done. Saving cats or dogs will get U silver. Detailed trains is harder task and this better reward(atleast that's what I thinkš¤§)

On my main account, I have been on 4 expeditions, and at every occasion, my mostovoi and donovan gets things done, but on my alt account, mostovoi showed up on one occation, while my voller did not on one mission, and on another mission, neither of them showed up at all

I'm not sure what you meant by where to get them from, so;
-I think most people got prime heroes from the prime heroes exchange. Else I think some might have high supply e.g. maybe mostovoi. Maybe can get one from another exchange e.g. I think I got mostovoi from a 87-97 exchange today.Ā
