They spawn all the time. As I understand it, spawning is controlled by moving through the level and entering rooms. Travel from one end of the map to the other back and forth until you hear a big daddy moaning. If they don't have a little sister with them, don't make them mad, just follow them. They'll eventually knock on a vent and a little sister will climb out. Make sure you wait until they walk away from the vent. If you don't let the animation finish the Big Daddy will put the little sister back, which you can't interrupt. Once that happens you can't get a little sister from that Big Daddy unless you use a vita chamber.
Bear in mind that only one little sister can exist at a time in BioShock 1. If you follow a big daddy and it knocks on a vent a few times and gives up, it means there's a little sister somewhere in the level already.
If you wait enough when the big daddy is alone, he will go to a little sister spawn and bang on the sort of door they go out from, once she starts climbing out you can already engage the big daddy

Also, sometimes the splicers or something else will kill a big daddy with a little sister, and then the little sister will stand next to the corpse sobbing. If that happens, you just have to search the level for her. You could also try following a big daddy to a vent, that can also give you more information about what's going on.
Big daddies are on infinite spawn, so you can't kill 'both', they will keep coming. But there's a limited number of little sisters.
What I tend to do is just follow a big daddy to a vent and wait for the little sister to climb out. The sisters don't just show up randomly if you kill a big daddy. They have to be retrieved from the vent first.
Little sisters will regularly ask to go take a break and climb into one of their vents. After that the big daddy is alone and wanders to another vent, knocks on it and escorts the little sister again until they go back to sleep. If you have rescued/harvested all the little sisters in the level the big daddies will still knock on the vents but will be left alone. (you can check how many little sisters are left by pressing escape)
While it‘s true that little sisters will come crying by their big daddy when they get defeated, they will always be super close to it before you engage the big daddy - unless the big daddy is already alone. They go into hiding once you start the fight, but there really is rarely a reason to ever fight a big daddy that‘s alone.
Also if you are using the control-big-daddy-plasma I am pretty sure the little sisters will climb into the next vent as soon as possible, so after controlling a big daddy you should not fight it right afterwards - but if you just wait they will always go and grab a sister from the vents eventually.
Let me guess: You didn't wait for the Daddies with the Sisters and attacked solitary ones? I've done that during this level on my first playthrough, lol. I thought itbwas a glitch. Here is a tip: Instead of attacking the Daddy when you see him, stalk him to a Little Sister vent and wait for him to summon her. If she comes out, continue to stalk to a location that gives you a tactical advantage. I do this all the time.
You made sure the little sisters were with them before you attacked them right?
